Lopez, 45, has shared the exciting news during Tuesday night's semi-final of "American Idol".
JLo will begin a Las Vegas residency at Planet Hollywood Resort & Casino in January 2016.
"I've been preparing for this moment my entire life," the singer and actress said in a statement, adding: "The show is going to be a multifaceted high energy Jenny from the Block party, mixing all aspects of what my fans and I love - Dance, Hip Hop, Latin and Pop. It's going to be the show I've always dreamed of creating."
Lopez is set to perform in 20 concerts at the resort's AXIS Theatre over the course of four months.
"Jennifer Lopez is the perfect addition to the exciting line-up at The AXIS," Planet Hollywood regional president David Hoenemeyer said in a statement.
Hoenemeyer's partner at Caesars Entertainment, Jason Gastwirth added: "Jennifer's show will be the perfect combination of a cutting-edge production spectacular and an extraordinarily fun night-out, all centred around a supremely talented international superstar."
